The Effect of Toothpaste Containing Chitosan Blangkas (Limulus Polyphemus) on the Growth of Streptococcus Mutans (In Vitro)

Introduction

Dental caries is a widespread condition caused by the bacterial activity of carbohydrates that can be fermented. Among the many bacteria involved, Streptococcus Mutans plays a significant role in the formation of dental caries. To combat this issue, various approaches can be applied, including mechanical, chemical, and immunological methods. One common way to prevent dental caries is to brush teeth twice a day and choose the right toothpaste. In this context, the use of toothpaste containing natural ingredients such as chitosan blangkas has gained attention due to its potential antibacterial properties.

Chitosan: A Natural Antibacterial Agent

Chitosan is a natural polymer obtained from the deacetylation of chitin, a polysaccharide found in the exoskeletons of anthropods, fungi, and yeast animals. Chitosan has been widely studied for its antibacterial, antifungal, and antiviral properties, making it a promising ingredient in the development of natural products for oral health. In this study, Blangkas chitosan was tested to evaluate its ability to inhibit the growth of Streptococcus Mutans.

Characterization of Blangkas Chitosan

The characterization of Blangkas chitosan revealed a degree of deacetylation of 84.20% and a molecular weight of 893,000 Dalton. These results indicate that the Blangkas chitosan used in this study has a high degree of deacetylation, which is essential for its antibacterial activity.

Research Purposes

The primary objective of this study was to compare the effectiveness of antibacterial toothpaste containing high-molecular chitosan with commercial toothpaste (sodium monofluorophosphate, NAMFP) and placebo toothpaste (without active substances). In this experiment, Streptococcus Mutans isolated from the USU FMIPA Biology laboratory was cultured on NA media.

Methodology

The antibacterial materials in this study were grouped into five categories:

Group I: High-molecular chitosan toothpaste (0.5%)
Group II: High-molecular Blangkas toothpaste (1.5%)
Group III: Chitosan toothpaste (2.5% high-molecular blanks)
Group IV: Commercial toothpaste (NAMFP) as a positive control
Group V: Toothpaste without active substances (placebo) as a negative control

After 24 hours of observation, the results showed that the groups using high-molecular Blangkas toothpaste in a concentration of 0.5% had the largest inhibitory zone for the growth of Streptococcus Mutans. This was followed by chitosan toothpaste groups at 1.5% and 2.5% concentrations. Meanwhile, commercial toothpaste (NAMFP) showed a smaller inhibition zone than other groups.

Results and Analysis

The analysis of one-way variance indicated a significant difference (p < 0.01) between toothpaste groups containing chitosan and other groups. Toothpaste containing high-molecular Blangkas chitosan has proven to be more effective in inhibiting the growth of Streptococcus Mutans compared to commercial toothpaste.

Conclusion

This study demonstrates that toothpaste containing high-molecular Blangkas chitosan can be a better alternative in preventing dental caries due to infection by Streptococcus Mutans. With a higher effectiveness, the use of toothpaste can provide additional protection for dental health. In the context of caries prevention, choosing products that contain natural ingredients such as chitosan Blangkas can be a more beneficial choice for dental and oral health.

The findings of this study have significant implications for the development of natural products for oral health.
