Introduction

The durian fruit, known for its unique smell and taste, has been a subject of interest in various fields of research. One of the most promising areas of study is the endophytic yeast population found within the fruit. Endophytic yeast are microorganisms that live within the plant tissues and have been shown to have various beneficial effects on plant growth and development. In this study, we investigated the potential of durian endophytic yeast in producing indol acetic acid (IAA) and dissolving phosphate.

Materials and Methods

To isolate and identify the endophytic yeast population, we used a combination of traditional and modern techniques. First, 25 grams of durian flesh were mixed with sterile distilled water until it reached a volume of 250 ml in a 1000 ml flask. This mixture was then diluted to a concentration of 10-15 cfu/ml using the Pour Plate method. The resulting yeast cultures were then incubated for 5 days at 29 ° C.

After incubation, each species of yeast was calculated, isolated, and morphologically identified in the MEA and biochemical media using the Vitex-2 Compact tool. The potential of each species of yeast in producing IAA was tested by growing 10 ml of inoculum in YMB (Yeast Malt Broth) media which contains 0.2 grams of L-tripopher. This culture was then incubated for 14 days at 29 ° C with a stirring speed of 110 rpm. IAA concentration measurements were carried out using a spectrophotometer at a wavelength of 520 nm.

To find out the potential of each species of yeast in dissolving phosphate, Pikovskaya media was used. The results showed that from the durian flesh there were five species of endophytic yeast, namely Brettanomyces Bruxellensis, Candida Ciferrii, C. Parapsilosis, Kloeckera Apiculata, and Pichia Anomala.

Results

The results of this study showed that all five species of endophytic yeast had the potential to produce IAA. Among them, Candida Ciferrii produced the highest concentration of 3.87 ppm, followed by Kloeckera Apiculata with 3.61 ppm. Furthermore, the potential for dissolving phosphate was also examined, where the Candida Ciferrii species showed the best ability with a clear zone of 9.60 mm and the phosphate solubility index reaches 1.03.

The findings of this study have significant implications for sustainable agriculture and the development of biotechnology. Utilization of endophytic yeast in plant cultivation can be an effective strategy to improve agricultural output, especially in improving soil fertility and increasing plant growth naturally.
