Sputum examination is carried out every 2 weeks for 2 months to monitor sputum conversion. The initial and end vitamin D levels are measured by the ELISA technique, while the polymorphism of the VDR gene is tested by the RFLP PCR technique.

The results showed that the intervention group that received vitamin D supplementation experienced sputum conversion faster than the comparison group (p <0.05). An increase in vitamin D levels is also far more significant in the intervention group (95.31%) than the comparison group (6.81%). Interestingly, the analysis of polymorphism APAI VDR gene shows that intervention groups with Homozygous AA genotypes and heterozygous AA experience sputum conversion faster in the 6th week, compared to intervention groups with Homozygous AA genotypes that experience conversion in the 8th week. However, the difference in sputum conversion time between the intervention group and the comparative group based on the APAI polymorphism is not significant (P> 0.05).

This study shows that the administration of vitamin D can accelerate sputum conversion in patients with Batak pulmonary TB. Polymorphism APAI VDR gene also shows the effect on sputum conversion time, where Homozygous AA genotypes and AA heterozygous in the intervention group experience faster conversion than Homozygous AA genotypes. This finding provides important information in handling pulmonary TB, especially in the Batak ethnic population.
