Introduction

Ovarian cancer is a malignant disease that attacks many women after menopause in Indonesia. The use of parp inhibitors is one of the therapeutic choices that are often used, but face the resistance constraints that often arise. This study aims to identify compounds that have the potential to be inhibitors of parp through pharmacofor modeling studies and test the results of identification based on lipinski rules, physicochemical properties parameters, and molecular docking.

The results of this study showed that based on the assessment of each ligand obtained from pharmacofor modeling and virtual filtering studies of Parp receptors (GDP code: 6NRJ), the compounds found were Zinc000012895188, Zinc000408664724, Zinc000197728633, Zinc000057895413, Zinc000408670550, Zinc 000299759100, Zinc000040156126, and Zinc000009046885.

Visualization of Amino Acid Residue Interactions

In visualization of amino acid residue interactions, zinc000012895188 compounds, zinc000057895413, zinc000040156126, and zinc000299817270 were found to have the potential to act as inhibitors parp. This is proven through the calculation of in silico through lower Gibbs (∆G) free energy in the tested ligands and the interaction of amino acid residues from the ligands that are tested similar to the comparison ligand, talazoparib.

Conclusion

This study opens opportunities for the development of new drugs based on parp inhibitors with higher effectiveness and lower resistance. Further development needs to be done through in vitro and in vivo tests to validate the results of this pharmacopho-modeling study and ensure the safety and effectiveness of these compounds as prospective parp inhibitors.

The results of this study suggest that pharmacofor modeling can be a useful tool in the identification of potential lead compounds for drug development.
