Introduction

The use of sodium nitrite as a preservative in processed meat products, including beef nuggets, has been a common practice in the food industry. Although its function is crucial in maintaining the quality and freshness of the product, the excessive and prolonged use of sodium nitrite can potentially have toxic effects on health. This study aims to analyze the level of sodium nitrite in beef nuggets circulating in the city of Medan, Indonesia, and ensure that the levels are in accordance with the limits set by the government.

Sodium nitrite is a widely used preservative in the food industry due to its ability to inhibit the growth of bacteria and extend the shelf life of meat products. However, its use has been linked to various health concerns, including cancer and cardiovascular disease. The World Health Organization (WHO) has classified processed meat, including beef nuggets, as a carcinogen, which has led to increased scrutiny of the use of sodium nitrite in the food industry.

In this study, we employed two methods to analyze the level of sodium nitrite in beef nuggets: qualitative and quantitative analysis. The qualitative method uses sulfanlic acid reagents and N-1-Naftylethylene-diamonium, which produces a purplish red color as an indicator of sodium nitrite. Meanwhile, the quantitative analysis was carried out using the UV-Vis spectrophotometry method at a wavelength of 545.50 nm to measure nitrite levels more accurately.

Materials and Methods

The study involved the analysis of three samples of beef nuggets, which were obtained from different sources in the city of Medan, Indonesia. The samples were labeled as Sample A, Sample B, and Sample C, and their expiration dates were January 31, 2018, January 24, 2018, and March 17, 2018, respectively.

The qualitative analysis was carried out using sulfanlic acid reagents and N-1-Naftylethylene-diamonium, which produces a purplish red color as an indicator of sodium nitrite. The reagents were added to the sample, and the resulting color was observed and recorded.

The quantitative analysis was carried out using the UV-Vis spectrophotometry method at a wavelength of 545.50 nm. The sample was placed in a cuvette, and the absorbance was measured using a UV-Vis spectrophotometer. The concentration of sodium nitrite was calculated using a calibration curve.

Results

The results of the study showed that the three samples of beef nuggets had varied sodium nitrite levels. Sample A had a nitrite content of 29.5 mg/kg, while Sample B had a nitrite content of 19.32 mg/kg. Sample C had the same nitrite content as Sample A, which was 29.5 mg/kg.
