Introduction

In the pharmaceutical world, paracetamol is one of the most commonly used drugs to treat pain and reduce fever. However, paracetamol has a low level of solubility in water, which often requires the use of co-solvents such as alcohol or propylene glycol to prepare a solution. The use of alcohol is not safe for children, while propylene glycol has been known to cause acute kidney failure in children. Therefore, suspension preparation is a solution to this problem, although the suspension also has weaknesses, namely the possibility of clumping. In this context, the authors are interested in developing paracetamol nanosuspension formulations that do not contain co-solvents and mixed solvents, and are able to prevent clumping.

Research Purposes

This study aims to make paracetamol nanosuspension using the top down method, characterize, and test the analgesic and antipyretic effects in vivo. In this study, the effects of nanosuspension will be compared with paracetamol suspension and solutions. The development of paracetamol nanosuspension is expected to provide a safer and more efficient formulation for patients, especially children. The use of nanosuspension technology can improve the solubility and bioavailability of paracetamol, making it more effective in managing pain and fever.

Methodology

This research was conducted through several stages. First, the identification of raw materials is done to determine the quality of the material to be used. Then, paracetamol nanosuspension is produced using the top down method. The top down method involves the use of high-energy milling to break down the paracetamol particles into smaller sizes. After manufacturing, the characterization of nanosuspensions is carried out, including organoleptic examination, measurement of particle size, potential zeta, pH, specific gravity, ratio, and solubility test and stability for three months. After the characterization stage, followed by testing therapeutic activity using white mice. This test includes analgesic tests using the hot plate method with five antipyretic treatment and tests carried out with induction of 2-6-dinitrophenol with six treatment groups.

Based on this research, it can be concluded that paracetamol can be formulated in the form of nanosuspensions using the top down method. Nanosuspensi has an advantage in terms of solubility that is faster compared to suspension. This discovery opens new opportunities in the development of drug formulations, especially to increase the effectiveness of paracetamol in the form of safer and efficient nanosuspension for patients, especially children. By understanding and applying this nanosuspension method, it is expected that the use of paracetamol can be more optimal, having a positive impact in managing pain and fever.
